abstract The present disclosure discloses targeting vectors, nucleic acid compositions and construction methods for constructing mouse models of liver injury, and relates to the technical fields of genetic engineering and genetic modification. A targeting vector of the present disclosure comprises a first expression cassette and a second expression cassette located downstream of the first expression cassette. The first expression cassette comprises in tandem a liver-specific promoter, a tetracycline transcriptional activation regulator, and a first polyA in sequence. The second expression cassette contains, in tandem, a second polyA, a gene encoding mouse urokinase-type plasminogen activator, and a tetracycline-inducible promoter, in sequence. A liver injury mouse model constructed with a targeting vector has a phenotype of spontaneous liver injury and exacerbation of induced liver injury, low mouse mortality in hybrid offspring, and can contribute to large-scale breeding. The present disclosure provides a reliable liver injury mouse model for liver disease research. [Selection drawing] Fig. 1
