This application is generally a polypeptide having specific target binding properties that is selectively delivered to a chimeric antigen receptor (CAR) composed of modified unnatural NKG2D receptors on genetically engineered mammalian cells. For example, it relates to the generation of a modified unnatural α1-α2 domain of an NKG2D ligand to which an antibody or variable fragment of an antibody is bound. Targeting of surface-expressed molecules included those of virus-infected cells, which subsequently expressed CAR homologous to the modified unnatural α1-α2 domain of the NKG2D ligand to which the polypeptide was bound. It is attacked and eliminated by genetically engineered cells of the immune system. [Selection diagram] None |
