The present invention generally relates to at least one cleavable moiety (CM1) and at least one serine protease (SP) or at least one cysteine protease (CP) which is a substrate for at least one matrix metalloprotease (MMP). A polypeptide containing at least a second cleavable moiety (CM2) as a substrate, at least one CM1 and at least one SP protease or at least one cysteine protease (CP) that is a substrate for at least one MMP protease. Activateable antibodies and other larger molecules containing these polypeptides, including at least one CM2 that is a substrate for at least one CM1 and at least one SP that is a substrate for at least one MMP protease. It relates to methods of producing these polypeptides comprising at least one CM2 which is a protease or a substrate for at least one cysteine protease (CP) and methods of using them in various therapeutic, diagnostic and prophylactic applications.
