abstract | Compositions and methods for treating and preventing Leber's hereditary optic neuropathy are provided. The present invention provides a method for treating and preventing LHON by providing a composition that promotes the production of ATP by mitochondria in the absence of NADH dehydrogenase activity. The composition comprises a water-soluble compound that is metabolized in the body to release a TCA intermediate, such as succinate. Since succinate does not rely on NADH dehydrogenase to contribute electrons to the mitochondrial electron transport chain, it drives mitochondrial ATP synthesis in individuals with mutations that cause LHON. Therefore, the methods of the invention compensate for metabolic deficiencies in such individuals to prevent or reduce retinal degeneration and vision loss. |
