abstract | An improved process for the preparation of tubular lysine compounds, tubular lysine drug linker compounds, and their intermediates is disclosed. The present invention provides a method for preparing a composition comprising a tubevalin compound in salt form, if desired, wherein the method is (a) contacting in a suitable polar aprotic solvent, (a). b) A step of forming an optical isomer mixture of tubevalin intermediates in salt form, or a composition containing this mixture, by quenching the reaction mixture with Bronsted acid, (c) the optical isomer mixture. The step of forming a composition containing an essentially equimolar mixture of diastereomers by contact with a suitable chiral reducing agent, (c') separating the diastereomers from the composition of the diastereomeric mixture. Including the step of obtaining a composition. |
