abstract | A method of detecting a target polynucleotide sequence in a given nucleic acid analyte: a. The analyte, and annealed to the single-stranded probe oligonucleotides A 0, at least partially double-stranded, and the 3 'end of A 0 to form an analyte target sequence duplex complex, the Produce one intermediate product; b. A first intermediate product, in the direction 3'-5 'from the 3' end of A 0, and pyrophosphorolysis in pyrophosphorolysis enzymes, to generate a partially digested strand A 1 and analyte C. (I) A 1 is annealed with a single-stranded trigger oligonucleotide B, and the A 1 strand is extended in the 5'-3'direction with respect to B; or (ii) at the 3'and 5'ends. A 1 is cyclized through ligation; or (iii) the 3'end of A 1 is ligated to the 5'end of the ligated probe oligonucleotide C; in each case, oligonucleotide A 2 is produced; d. Primed with at least one single-stranded primer oligonucleotides A 2, and generates a large number of copies or A 2 region of A 2; and e. The method characterized by the steps of detecting a signal from multiple copies and inferring from it the presence or absence of a polynucleotide target sequence in the analyte. |
