abstract |
The present disclosure provides a method for identifying bisulfite-free in nucleic acid sequences at the positions of 5-methylcytosine, 5-hydroxymethylcytosine, 5-carboxylcytosine and 5-formylcytosine. The methods described herein provide DNA or RNA methylation and hydroxymethylation analysis with a mild reaction that quantitatively detects modified cytosines at base resolution without affecting unmodified cytosines. Combining TET oxidation and reduction with borane derivatives (eg, pyridineborane and 2-picolinborane (pic-BH 3)), referred to herein as TAPS (TET Assisted Pyridine borane Sequanting).
