abstract PROBLEM TO BE SOLVED: To provide a new diabetes treatment technique which reduces the risk of hypoglycemia but does not deviate upward from the blood glucose concentration beyond the normal range. SOLUTION: (i) A chain modified with a monomeric glucose-binding element at or near its N-terminal, and (ii) reversible to a monomeric glucose-binding element so that this bond can be replaced by glucose. Disclosed is a double-stranded insulin analog containing a modified B chain at or near its C-terminal by a ligated element.
