A kit for measuring pneumoniae mycoplasma nucleic acid and drug resistance gene mutations thereof, which comprises a KOD DNA polymerase, a primer pair of a specific sequence, and a specific target fluorescent probe. By designing primers and probes of specific sequences, mutations at the 2063 or 2064 locus of pneumoniae mycoplasma nucleic acid, pneumoniae mycoplasma nucleic acid 23S rRNA gene and 23S rRNA can be detected quickly, easily and accurately.
