PROBLEM TO BE SOLVED: To provide a method, application and system for obtaining the most sensitive data on rare circulating tumor cells (CTC) in a blood sample. A method of analyzing circulating tumor cells (CTCs) in a blood sample, wherein a product containing CTC and other cells is isolated from the blood sample, and the volume of the product is reduced. Isolating ribonucleic acid (RNA) molecules from the product, generating cDNA molecules from isolated RNA in solution, encapsulating the cDNA molecules in individual droplets, and one or more reporters. Amplifying the cDNA molecule in each droplet in the presence of a group, detecting the droplet containing the reporter group as an index of the presence of the CTC-derived cDNA molecule in the droplet, and detecting the liquid.
