PROBLEM TO BE SOLVED: To provide a composition, a method and a system for sequencing a template nucleic acid by using a polymerase-based sequencing procedure by binding. A scrutiny step involves monitoring the interaction of the polymerase with a template nucleic acid in the presence of one or more nucleotides. The identity of the next proper nucleotide in the sequence is determined without the incorporation of the nucleotide into the primer structure by the formation of phosphodiester bonds. The optional integration step can be used after a scrutiny step in which the primer is extended with one or more nucleotides, thereby incrementing the template nucleotides that can be scrutinized in subsequent scrutiny steps. The binding sequencing procedure does not require the use of labeled nucleotides or polymerases, but these reagents can be optionally utilized. [Selection diagram] None |
