abstract | PROBLEM TO BE SOLVED: To develop a method for enzymatically synthesizing a (oligo) peptide. SOLUTION: The method of the present invention comprises coupling (a) a (oligo) peptide C-terminal ester or thioester with (b) an (oligo) peptide nucleophilic reagent having an N-terminal unprotected amine, the coupling comprising water. Performed in the containing fluid, the coupling was performed as compared to the subtilisin BPN' or its homologous sequence represented by SEQ ID NO: 2, the following mutations: -Amino acid deletion corresponding to positions 75-83,- By a mutation at the amino acid position corresponding to S221, a mutation that is S221C or S221 selenocysteine-preferably by a subtilisin BPN'mutant or a homologue thereof, including a mutation at the amino acid position corresponding to P225.
