PROBLEM TO BE SOLVED: To provide an alternative method and a compound used for treating Alzheimer's disease and diagnosing or treating a tumor. A method for producing a mutant protein derived from human lipocalin 2 (Lcn2, hNGAL), which can bind to a predetermined unnatural target having a detectable affinity and encodes a human Lcn2 protein. One nucleic acid molecule is mutated with a nucleotide triplet encoding at least one of the sequence positions corresponding to sequence positions 96, 100, and 106 in the linear polypeptide sequence of human Lcn2. A method comprising step (a) of producing or more mutant protein nucleic acid molecules. Use of a mutant protein derived from the Lcn2 protein according to the production method, as well as a mutant protein used for the treatment or diagnosis of Alzheimer's disease.
