PROBLEM TO BE SOLVED: To provide a genetically engineered CRISPR-Cas9 nuclease having improved specificity. A variant of Cas9 protein with reduced binding affinity for Cas9 protein with increased specificity compared to wild-type proteins (ie, substantial at incompletely matched or mismatched DNA sites). Numerous Cas9 variants (which induce less off-target effects), and their use in genomic genetic engineering, epigenetic genetic engineering, genomic targeting, and genome editing.
