abstract | PROBLEM TO BE SOLVED: To provide a novel positively charged prodrug of a peptide and related compounds. SOLUTION: A compound (structure 1) that can be synthesized by a standard peptide synthesis protocol. [Effect] The positively charged amino group of the peptide prodrug not only dissolves the drug in water, but also binds to the negative charge on the phosphate head group of the membrane, pushing the prodrug into the cytosol. Prodrugs diffuse through human skin 100 to 1000 times faster than peptides and related compounds. Prodrugs can be used to treat peptides and related compounds in human or animal therapeutic conditions. Prodrugs can be administered transdermally for any type of drug treatment and can avoid proteolytic enzyme degradation in the gastrointestinal tract. [Selection diagram] Fig. 2 |
