Some aspects of the present disclosure provide strategies, systems, reagents, methods, and kits that are useful for targeted editing of nucleic acids, including the ability to identify single sites in the genome of a cell or subject, such as in the human genome. Including editing. In some embodiments, a fusion protein of a nucleic acid programmable DNA binding protein (napDNAbp), such as Cpf1 or a variant thereof, and a nucleic acid editing protein or protein domain, such as a deaminase domain is provided. In some embodiments, methods for targeted nucleic acid editing are provided. In some embodiments, for the creation of a fusion protein of a nucleic acid editing protein or domain with a targeted nucleic acid editing protein, such as napDNAbp (eg, CasX, CasY, Cpf1, C2c1, C2c2, C2c3, and Argonaute).
