Methods and compositions are provided for treating or preventing cognitive impairment in a mammal, preferably a human. The method generally involves the activation of a GPR158 signaling pathway that includes osteocalcin, for example, by administration of undercarboxylated / uncarboxylated osteocalcin. Disorders treatable by the method include cognitive deficits due to age-related neurodegeneration, anxiety, depression, memory loss, learning disabilities, and cognitive deficits associated with food shortages during pregnancy.
