A genome editing system is provided for site-specifically modifying a target sequence in the genome of a cell, comprising an expression construct comprising both the coding sequence and the coding sequence for a tRNA-gRNA or ribozyme-gRNA. Also provided are methods for genetically modifying cells by introducing a genome editing system with high efficiency and high specificity.
