abstract | The present invention provides a novel method for labeling a nucleic acid probe. The method uses a ligase-catalyzed reaction to link a nucleic acid probe and a previously prepared labeled carrier molecule of a nucleic acid in the presence of a stabilizing complementary splint oligonucleotide. The method allows multiple probes to be easily, inexpensively, and quickly labeled with multiple different labels. Thus, the cost and effort for creating a single molecule fluorescence in situ hybridization (smFISH) assay is greatly reduced.
