abstract |
PROBLEM TO BE SOLVED: To make a human epitope-binding protein containing a common light chain without relying on manipulating in vitro conditions, and instead using a more biologically reasonable method. The composition and method are provided. A genetically modified mouse expressing a human immunoglobulin heavy chain and a light chain variable domain, the mouse having a limited light chain variable repertoire; binding to a diverse repertoire of affinity matured human heavy chain variable domains. And a biological system for producing a human light chain variable domain that is expressed together; a method for producing a binding protein containing an immunoglobulin variable domain, the purpose of which is a mouse having a limited immunoglobulin light chain repertoire. There is provided a method comprising immunizing with an antigen of the above and using a mouse immunoglobulin variable region gene sequence for a binding protein that specifically binds to the antigen of interest.
