abstract |
PROBLEM TO BE SOLVED: To provide a method for in vitro inducing a glandular pituitary gland or a precursor tissue thereof from human pluripotent stem cells. SOLUTION: Agglomerates of human pluripotent stem cells are suspended and cultured in a medium containing an osteogenic factor signal transduction pathway activator and a Shh signal pathway activator to obtain hypothalamic neuroepithelial tissue and epidermal ectoderm. Human cell agglomerates containing, and further obtained human cell agglomerates containing hypothalamic neuroepithelial tissue and epidermal ectoderm in a medium containing an osteogenic factor signaling pathway activator and a Shh signaling pathway activator. Further suspension culture induces the formation of pituitary placode and / or Rathke's pouch in the epidermal ectoderm, 1) hypothalamic neuroepithelial tissue, and 2) humans containing pituitary placode and / or Rathke's pouch.
