The preparation of aminoalcohol lipidoids by reacting an amine with an epoxide-terminated compound is described. Methods for preparing amino alcohol lipidoids from commercially available starting materials are also provided. Amino alcohol lipidoids can be prepared from racemic or stereochemically pure epoxides. The aminoalcohol lipidoid or salt forms thereof are preferably biodegradable and biocompatible and can be used in various drug delivery systems. Due to the amino moiety of these amino alcohol lipidoid compounds, they are particularly suitable for polynucleotide delivery. Complexes, micelles, liposomes or particles containing the lipidoids and polynucleotides of the invention were prepared. The lipidoids of the present invention can also be used in the preparation of microparticles for drug delivery. They are particularly useful for the delivery of labile agents due to their ability to buffer the pH of their surroundings. [Selection diagram] None |
