abstract A method for preparing a purified polypeptide composition is provided. The present invention relates to purified peptidomimetic macrocycles. The invention further provides methods of preparing and using such macrocycles, for example, in therapeutic applications. In one aspect, the invention provides a method for preparing a purified peptidomimetic macrocycle composition. In one embodiment, the method comprises contacting the peptidomimetic precursor and purifying the crude peptidomimetic macrocycle to obtain a purified peptidomimetic macrocycle composition. In such embodiments, the peptidomimetic precursor may include at least two moieties that can undergo a metathesis reaction using a metathesis catalyst to produce a crude peptidomimetic macrocycle.
