abstract |
The present invention relates to methods and compositions for providing an environment for promoting improved protein expression and / or folding. More specifically, the present invention relates to a recombinant thermostable ferritin assembly comprising at least one genetically modified ferritin subunit, wherein the at least one genetically modified ferritin subunit is a wild type ferritin subunit. It lacks the unstructured carboxy-terminal sequence present in the unit, and the recombinant ferritin assembly is more stable than wild-type ferritin assembly at low salt concentrations A recombinant ferritin assembly is provided. The present invention is also exemplified by a ferritin assembly derived from the hyperthermophilic bacterium Archaeoglobus fulgidus. Further claimed are nucleic acids, compositions, kits and methods for encapsulating a polypeptide in the recombinant ferritin assembly and delivering and administering the ferritin assembly to a subject. |
