abstract | Described herein are methods and compositions related to the generation of induced pluripotent stem cells (iPSCs). An improved technique for establishing highly efficient and reproducible reprogramming using non-integrated episomal plasmid vectors is described. Using the described reprogramming protocol, non-T cells can be consistently reprogrammed with near 100% success rate from non-T cell or non-B cell sources. Further advantages include the use of a defined reprogramming medium E7 and the use of recombinant human L-521, a defined clinically compatible substrate. Generation of iPSCs from these blood cell sources enables iterative generation of the entire genome repertoire, preservation of genome fidelity and enhanced genome stability. [Selection] Figure 3 |
