Disclosed herein are systems and methods for transducing, activating, or otherwise processing cells. Cells are introduced into the inner layer of a multilayer stack that defines at least one flow chamber and a plurality of cell-associated regions. Vertical flow through the stack allows cells to be entrained in the cell-associated area with genetic information transfer agents or other additives before the cells are washed with reverse vertical flow and withdrawn from the device.
