abstract | The present disclosure presents an additive system for use in PEGylation of proteins. The additive system includes p-aminobenzoic acid hydrazide used alone or in combination with an aromatic amine such as 3,5-diaminobenzoic acid or an ammonium salt such as ammonium chloride or ammonium acetate. The combination of additives of the present disclosure provides several benefits such as increased reaction rate, high yield, and reduced aminooxy-PEG equivalents required to compete for the coupling reaction. A typical reaction can proceed by combining an additive or additive system with a solution of protein and aminooxy-PEG reagent. The solution is adjusted to pH 4 and kept at 20-25 ° C. without stirring until completion, generally within 24 hours. [Selection] Figure 6 |
