abstract | The present invention relates to a Neisseria LPS having a tetraacylated lipid A moiety, wherein the tetraacylated lipid A moiety lacks one of the secondary acyl chains compared to the lipid A moiety of wild type Neisseria LPS; And is modified in that it lacks the primary acyl chain at position 3 of glucosamine located at the reducing end of the lipid A moiety. The invention further relates to a Neisseria bacterium that has been genetically modified to reduce the expression of one of the endogenous lpxL1 or lpxL2 genes and introduce the expression of a heterologous pagL gene. The Neisseria LPS of the present invention has TLR4 agonist properties and is therefore useful in compositions that elicit or stimulate an immune response, such as a vaccine, as well as other forms of immunotherapy.
