abstract |
A recombinase mutant for improving recombinase-mediated amplification, such as a PCR library having a single-stranded adapter region, is provided. A variant of UvsX recombinase comprising an amino acid sequence at least 90% identical to the enteric bacterial phage RB49 UvsX recombinase having a specific amino acid sequence, wherein the variant is functionally equivalent to position 256 of said RB49 UvsX recombinase Excluding the substitution of His to Ser at position 63 of the UvsX recombinase of enterobacterial phage T4 containing a substitution mutation to a charged amino acid residue at a position and having a specific sequence, or the position 63 of the RB49 UvsX recombinase. A variant of UvsX recombinase having increased recombinase activity as compared to a variant enterobacterial phage RB49 UvsX recombinase having the entire amino acid sequence of said RB49 UvsX recombinase.
