Synthetic combinatorial AAV capsid library for targeted gene therapy |
abstract |
The various strategies for generating capsid libraries that have been developed so far all suffer from sequence bias or limited diversity. SOLUTION: For the production of modified adeno-associated virus (AAV) cap genes and for the production of chimeric AAV vectors and combinatorial libraries of virions, for the selection of virions displaying cell specific affinity and for specific implementations In form, compositions and methods are disclosed for the production of helper vectors containing one or more modified AAV cap genes. The synthetic combinatorial AAV capsid libraries of the present invention are useful for the introduction of one or more nucleic acid molecules into selected target host cells. The viral vectors and gene constructs disclosed herein are also useful for various gene therapy diagnostics and / or treatment regimens. [Selection] Figure 2 |
