Disclosed are a deletion mutant of Cas9 protein and a method for producing chimeric Cas9 protein. A part of the protein has been modified to be deleted, but it functions as an RNA-induced DNA-binding nuclease that can bind to the target DNA and form a double-strand break in the target DNA, Functions as an RNA-induced DNA binding nickase that binds to DNA and is guided by one or more types of RNA, or can bind to target DNA and form a single-stranded break or nick in target DNA; Cas9 protein which is an RNA-induced DNA binding protein of type II CRISPR system.
