abstract The present invention discloses a method and reagent kit for measuring the biological activity of methionine adenosine transferase (MAT). A MAT-catalyzed enzymatic reaction occurs by mixing a sample and a reagent in a fixed ratio, and a competitive enzyme-linked immunosorbent assay for measuring this reaction and its product S-adenosylmethionine (SAM); It can be carried out stepwise or simultaneously. The MAT biological activity was calculated by measuring the absorbance of the reaction product of the tracer enzyme in a specific spectrum and calculating the amount of product SAM produced per unit time compared to the absorbance of the standard product. In the method of quantifying SAM, the tracer is labeled with an anti-SAM antibody or SAM or SAM analog antigen, and the produced SAM binds to the anti-SAM antibody competitively with a certain amount of SAM antigen by the competition principle. A kit using the method of the invention makes the determination of MAT activity more sensitive, accurate, reliable, direct, simple and quick. The method of the present invention was used to measure MAT activity in normal and liver cancer cells and MAT activity under the influence of different regulators. [Selection] Figure 3
