abstract |
The present invention relates to a retroviral producer cell comprising a gag and pol protein; an envelope protein or functional replacement thereof; and a nucleic acid sequence encoding the RNA genome of a retroviral vector particle, said nucleic acid sequences all comprising retroviral production. Located at a single locus in the cell genome. The invention also provides a nucleic acid vector comprising a non-mammalian origin of replication and the ability to retain at least 25 kilobases (kb) of DNA, comprising gag and pol proteins, and env proteins or functional substitutions thereof. It relates to a nucleic acid vector characterized in that it comprises a coding retroviral nucleic acid sequence. The invention also relates to the use and methods of use of said nucleic acid vectors for producing stable retroviral packaging and production cell lines. |
