abstract Embodiments of the present disclosure relate to the use of expression constructs, in which at least one poly A signal is embedded upstream of the expressible transcript, eg, within the 5 ′ UTR of the transcript. In certain embodiments, a poly A signal is included in the ligand binding aptamer and the ligand binding to the aptamer or absence of binding indicates the outcome of the expressible transcript. In certain embodiments, in the absence of a ligand, an expression transcript having poly A in the 5′UTR is expressed but subsequently degraded, while the presence of the ligand is degraded when an expressible transcript is expressed. Inhibits. Multiple ligand binding aptamers can be present on the same expression construct. [Selection] Figure 1A
