abstract | The present disclosure relates generally to modified Gram-positive bacteria cells that produce increased amounts of one or more proteins of interest and modified Gram-positive bacteria cells that have increased gene competency. Accordingly, certain embodiments of the present disclosure relate to modified Gram-positive bacterial cells that express increased amounts of the target protein relative to unmodified (parent) Gram-positive bacterial cells that express the same protein of interest, wherein the modified bacterial cell Contains at least one mutation in the rpoC gene encoding a mutant RNA-polymerase (RNAP) β′-subunit polypeptide. In certain embodiments, the rpoC gene encoding the mutant β′-subunit polypeptide is integrated into the chromosome of the modified cell. In other embodiments, the rpoC gene encoding the mutant β′-subunit polypeptide is contained on an extrachromosomal plasmid introduced into the modified cell. In other embodiments, the disclosure provides at least one nucleic acid construct encoding a modified rpoC polypeptide comprising 90% sequence identity to SEQ ID NO: 8 and a substitution of aspartate to glycine at position 796 of SEQ ID NO: 8. A competent Bacillus host cell comprising a copy, wherein a polynucleotide encoding an rpoC polypeptide is non-competent prior to introduction of the first nucleic acid construct and a Bacillus host cell Is exogenous.
