abstract |
The present disclosure uses a nuclease that has substantially higher activity on double-stranded DNA compared to single-stranded DNA or single-stranded RNA and targets for subsequent enrichment relative to wild-type nucleic acid. Methods are provided for preparing mutant nucleic acids. The present disclosure also includes a method for enriching the target mutant nucleic acid and for preparing an unmethylated / methylated nucleic acid of interest for subsequent enrichment. This method is a novel method that results in preferential cleavage of the wild-type nucleic acid, thereby allowing subsequent enrichment of the mutated target sequence of interest. |
