Described herein is a method for producing a chimeric non-human animal that expresses the human ETV2 gene, comprising the following steps: (a) generating an ETV2 null non-human animal cell in which both copies of the non-human ETV2 gene have a mutation that interferes with the production of functional ETV2 protein in the non-human animal; (b) Fusing the nucleus derived from the ETV2 null non-human animal cell of (a) to an enucleated non-human oocyte, and activating the oocyte to produce an ETV2 null non-human blastocyst Creating an ETV2 null non-human blastocyst by somatic cell nuclear transfer, including dividing to form; (c) introducing human stem cells into the ETV2 null non-human blastocyst of (b); and (d) Transplanting the blastocyst of (c) into a surrogate mother non-human animal during pseudopregnancy to produce a chimeric non-human animal expressing human ETV2. |
