abstract | Provided herein are methods and compositions for depleting targeted nucleic acid sequences from a sample, enriching for a sequence of interest from a sample, and / or splitting a sequence from a sample. In one aspect, a method for enriching a sample for a sequence of interest comprising: (a) providing a sample comprising a sequence of interest and a targeting sequence for depletion, wherein the sequence of interest comprises: Constituting less than 30%; (b) contacting the sample with a plurality of CRISPR / Cas system protein-gRNA complexes, wherein the gRNA is complementary to the targeting sequence and thereby targeted A method is provided herein comprising the step of cleaving the sequence. In any of the embodiments disclosed herein, the method further comprises extracting a sequence of interest and a targeting sequence for depletion from the sample. |
