abstract |
The present invention provides a double-stranded nucleic acid molecule comprising a 5 ′ extension of the sense strand or antisense strand and further comprising a plurality of nucleotides conjugated to a ligand, and methods of using the double-stranded nucleic acid molecule. Ligand-modified oligomers in which the sense strand forms a tetraloop provide a powerful and stable novel RNA interference agent. These dsNA molecules are synthesized using a plurality of nucleotides including ligand modified monomers, nucleotide analog monomers, modified nucleotide monomers, etc. using standard nucleotide synthesis methods and systems. |
