Recovery of visual response by in vivo delivery of rhodopsin diffusion |
abstract |
PROBLEM TO BE SOLVED: To provide recovery of visual response by in vivo delivery of rhodopsin diffusion. A nucleic acid vector encoding a light-gated cation-selective membrane channel, in particular channelrhodopsin-2 (Chop2), is used in an animal model to sensitize inner retinal neurons to light-sensitive cells in a photoreceptor degenerated retina. Converted to. Such treatment restored various aspects of visual cognition and vision. A method of restoring photosensitivity to the retina of a subject who is blind due to photoreceptor degeneration is provided as well as in retinitis pigmentosa or macular degeneration. This method comprises the above-described nucleic acid vector comprising an open reading frame encoding rhodopsin, wherein the promoter and transcriptional regulatory sequences are operably linked to the subject by intravitreal or subretinal injection. And delivering a vector whose nucleic acid is expressed in internal retinal neurons. [Selection] Figure 7 |
