A method of partially oxidizing a carbon source to generate a gas containing carbon monoxide, and causing a microorganism to generate an organic substance from the gas in a fermenter, and in the partial oxidation of the carbon source, rather than air A method in which a step of generating an organic substance by supplying a gas having a high oxygen concentration is performed under a condition that utilization is 0.5 or more, preferably 0.7 or more. In the partial oxidation of the carbon source, the pressure of the gas in the fermenter is preferably 1.17 bar-abs or more, preferably 1.69 bar-abs or more. [Selection] Figure 1
