The present invention provides a new direct tool for high-resolution structural analysis of individual conformers of G protein-coupled receptors (GPCRs). Contacting the Nanobody with the GPCR, and an agonist of the GPCR, and improving the binding of the Nanobody to the GPCR in the presence of the agonist compared to the GPCR in the absence of the agonist. Detecting the enhanced affinity of the GPCR to the agonist in the presence of the Nanobody compared to the GPCR in the absence of the Nanobody, wherein the GPCR is in an active conformational state.
