abstract Provided is a nucleic acid amplification reaction method capable of detecting nucleic acid amplification with high sensitivity even when PCR is accelerated. The method includes a step of applying a thermal cycle for amplifying the nucleic acid to a reaction solution containing a nucleic acid amplification reaction reagent used for the amplification of nucleic acid, and for annealing reaction and extension reaction in the thermal cycle. The heating time is 1 second or more and 10 seconds or less, and the nucleic acid amplification reaction reagent includes a forward primer, a reverse primer, a polymerase, and a fluorescently labeled probe, and the concentration of the forward primer is 0.4 μM or more and 3.2 μM or less. The concentration of the reverse primer is 0.4 μM or more and 3.2 μM or less, the concentration of the polymerase is 0.5 U or more and 4 U or less, and the concentration of the fluorescently labeled probe is 0.15 μM or more and 1.
