abstract Nucleic acid sequencing using nanopore detection is provided. An MspA mutant which is a porin derived from Mycobacterium smegmatis, wherein a specific 37 amino acids of MspA are replaced with specific amino acids at each amino acid position. Msp variants improve nucleotide discrimination. In addition, the mutant increases the current range (which makes it easier to distinguish different nucleotides) and reduces state fluctuations (which increases the signal-to-noise ratio). In addition, as the nucleic acid moves through the pore, the number of nucleotides involved in the current decreases. This makes it easier to identify the direct relationship between the current observed when the nucleic acid moves through the pore and the nucleic acid sequence. [Selection figure] None
