abstract The present invention provides a composition for specific inhibition of gene expression by dsRNA having modifications. An isolated double-stranded RNA (dsRNA) comprising: i) a sense strand having a length of 19 to 80 nucleotides; and ii) an antisense strand having a length of 19 to 35 nucleotides. The sense strand and the antisense strand form a duplex of 15-35 base pairs in length; the sense strand includes a tetraloop adjacent to the duplex, and the tetraloop is 2 ′ -Containing at least one nucleotide with a sugar substitution; said dsRNA comprising a discontinuity between the 5 'end of said sense strand and the 3' end of said antisense strand; and said dsRNA in mammalian cells An isolated dsRNA that reduces target gene expression. The tetraloop is a dsRNA having a nucleic acid sequence selected from UNCG, GNRA, CUUG, d (GNNA), d (GNAB), d (CNNG) and d (TNCG). [Selection] Figure 1A
