abstract |
Provided is a gene therapy method for the treatment of pathogen infection using a rationally designed LAGLIDADG meganuclease. A method for treating a human disease by gene therapy in a eukaryotic cell using a recombinant meganuclease having altered specificity for at least one recognition sequence half site compared to wild-type I-CREI meganuclease. Wherein said recombinant meganuclease comprises a polypeptide having at least 85% sequence similarity to residue 2-153 of I-CREI meganuclease and of a specific sequence within the I-CREI meganuclease recognition sequence A method comprising a modification that has specificity for a half site and a recognition sequence half site that differs by at least one base pair.
