abstract A method for purifying a target nucleic acid is provided. The present invention relates to a method for purifying at least a target nucleic acid from a sample, the method comprising at least a) incubating the sample with at least one proteolytic compound; and b) a target nucleic acid. Binding to the solid phase; c) eluting the target nucleic acid from the solid phase; d) incubating the eluted target nucleic acid with at least one proteolytic compound; e) re-immobilizing the target nucleic acid. Binding to the phase; and f) optionally eluting the bound target nucleic acid from the solid phase. Surprisingly, performing the second protein digestion step after the target nucleic acid is bound and eluted from the solid phase but before the nucleic acid is rebound to the solid phase remains in the isolated nucleic acid. It has been found to be very efficient in reducing protein contaminants. [Selection figure] None
