abstract The present disclosure provides systems, processes, products and compositions relating to the use of degradable adapters for background reduction in various nucleic acid manipulations. In particular, adapters are provided that can be degraded to the extent that degradation products cannot participate in, or substantially cannot, participate in subsequent reactions (eg, ligation, primer extension, amplification and sequencing reactions). The degradable adapter can be a partially double-stranded oligonucleotide adapter, a single-stranded oligonucleotide adapter, a stem-loop oligonucleotide adapter, or any type of oligo that can form a dimer by ligation and / or primer extension. It can be a nucleotide adapter.
