abstract |
This invention relates to an in vitro method for detecting class II restricted CD4 + T cells in a sample. Here, the sample is brought into contact with the separated complex of MHC class II molecules and peptides. This peptide is a [CST] -xx-C or C-xx- [CST] motif, separated from the MHC class II restricted T cell epitope of the antigen protein by a linker that is immediately adjacent to it or up to 7 amino acids. And a sequence having CD4 + T cells are detected by measuring the binding of the complex with cells in the sample, and the binding of the complex to the cells indicates the presence of CD4 + T cells in the sample. The invention is further a separate complex of an MCH class II molecule and a peptide, wherein the peptide is an MHC class II restricted T cell epitope of an antigenic protein and a linker of up to 7 amino acids immediately adjacent to it.
