abstract |
Disclosed herein are methods, compositions, and kits for highly efficient site-specific genome editing of cells. A method for altering a target polynucleotide sequence in a cell comprising the regularly spaced short palindromic repeat-associated (Cas) protein clustered with the polynucleotide sequence and one to two ribonucleic acids A cell in which the ribonucleic acid directs the Cas protein to the target motif of the target polynucleotide sequence and hybridizes with the target motif, wherein the target polynucleotide sequence is cleaved and expresses the Cas protein Wherein the efficiency of change is from about 50% to about 80%. |
